A hybridisation beacon which is an oligonucleotide where (a) the oligonucleotide has no secondary structure arising from one region of the oligonucleotide which hybridizes with another, (b) the oligonucleotide is formed of nucleotide residues of which one is labeled with a reporter fluorescent wherein the beacon includes a single indicator without a quencher associated and the indicator is selected from the group of 6-carboxyfluorescein, tetrachlorofluorescein and hexachlorofluorescein, (c) the labeled nucleotide residue indicator is positioned internally within the oligonucleotide sequence, ( d) the oligonucleotide is fully complementary to one allele of human genomic DNA having a polymorphism known in the binding site of the oligonucleotide, and (e) the oligonucleotide probe is modified at its 3 '' as to prevent chain extension during PCR amplification.
