摘要

The invention relates to a fusion protein called GST-fucosidase (GST-FUC), which exhibits alpha-L-fucosidase enzymatic activity, produced in the Escherichia coli bacteria by expression of the recombinant plasmid pGEX-FUC. This plasmid includes a cDNA sequence encoding the human alpha-L-fucosidase enzyme inserted into the BamHI site of the prokaryotic expression vector pGEX-2T, which includes the DNA sequence encoding the glutathione S-transferase protein (GST) of the helminth Schistosoma japonicum. The GST-FUC protein, purified by affinity chromatography, is digested with the thrombin protease to obtain the recombinant human alpha-L-fucosidase protein. These recombinant proteins can be used for the preparation of polyclonal and monoclonal antibodies, as well as for structural studies.
