RECOMBINANT PROTEINS CONTAINING LIPID-BINDING DOMAINS AS SERUM-FREE CELL CULTURE MEDIUM SUPPLEMENTS FOR ENHANCED BACULOVIRUS INFECTION, BACULOVIRUS PRODUCTION AND PRODUCTION OF RECOMBINANT PROTEINS PRODUCED BY BACULOVIRUS EXPRESSION VECTORS: GENERATION AND METHODS OF USE
Described herein is a method for achieving high rates of entrance into and generating high titers of beculoviruses by lepidopteran cells maintained in serum-free culture media. The method is based on the generation and use of transformed lepidopteran cell lines that are maintained in serum-free media and constitutively over-express a secreted factor(s) containing a lipid-blinding, MD2-related lipid recognition (ML) domain. The transformed cell lines grown in serum-free media supplemented withconditioned media obtained from the transformed lines display enhanced rate of infection by baculoviruses and produce higher numbers of baculovirus progeny than untransformed cells grown in non-supplemented serum - free media.
