Free kappa- and/or free lambda immunoglobulin light chains are detected quantitatively in the cerebrospinal fluid of the patient. Detection follows addition of a specific antibody, and employs latex-assisted nephelometry, turbidimetry or radio-immuno assay of the cerebro-spinal fluid, for quantitative measurement. In addition, light chains are also measured in the serum. Increase of free kappa immunoglobulin light chains in the cerebro-spinal fluid, indicates an auto-immune process. Increase in the free lambda immunoglobulin light chains indicates meningitis caused by a pathogen. Conditions which may be indicated in accordance with the foregoing principles are further discussed. They include the advance of multiple sclerosis, meningitis conditions and viral or bacterial conditions e.g. associated with borrelia, or syphilis. A test strip is proposed for rapid diagnosis of meningitis initially suggested by headache symptoms. Corresponding methods of investigation are also claimed.
