PROBLEM TO BE SOLVED: To provide a method for producing a heat-resistant firefly luciferase by changing specific amino acid residues in a mild type firefly luciferase into hydrophobic amino acid residues.;SOLUTION: This heat-resistant firefly luciferase is characterized by changing the amino acids at the 217- and 315-positions in the amino acid sequence of the mild type firefly luciferase or the amino acids at the positions equivalent to the 217- and 315-positions in the amino acid sequence of Luciola cruciata or Luciola lateralis luciferase into hydrophobic acid residues.
