PURPOSE: A direct electrical detection method of DNA-hybridization and a DNA chip fabrication method are provided, thereby requiring no expensive detecting substance and device, reducing the number of the detection process, and improving detecting accuracy and reliability. CONSTITUTION: The direct electrical detection method of DNA-hybridization comprises the steps of: (a) introducing deionized water or gas into the surrounding of the probe DNA(18) in a DNA chip of which both ends are fixed to each electrode through a DIV/gas inlet-hole(14), or subjecting the probe DNA to vacuum condition, and supplying electricity to the probe DNA to measure electric value of the single strand probe DNA; (b) reacting the both ends-fixed or one end-fixed probe DNA with a sample DNA in buffer solution to induce hybridization; and (c) removing unreacted sample and buffer solution from the DNA chip, introducing deionized water or gas into the surrounding of DNA, or subjecting DNA to vacuum condition, and supplying electricity to DNA to measure electric value of each DNA, and comparing the value of (c) with the value of (a) to decide occurrence of hybridization. The method for fabrication of the DNA chip comprises the steps of: (a) forming micro- or nano-channels(19) on a substrate(11); (b) forming electrodes in the channels; and (c) fixing both ends of the probe DNA(18) to each positive electrode(12) and negative electrode(13).
