A process for large scale purification of a ciclosporin, rapamycin or ascomycin from impurities with closely-related physical properties comprises counter-current extraction using heptane-acetone or heptane-isopropanol and water-acetone or water-isopropanol - DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for: - (1) a countercurrent extraction column with 100 to 200 compartments and an overall efficiency of 10 to 30%; - (2) a bulk quantity of ciclosporin A with and impurity level of less than 0.5% by area using HPLC; and - (3) compositions comprising the high purity ciclosporin A.
