Method for simultaneously determining fluorescence emissions from two fluorochromes (F1; F2) having overlapping emission wavelengths using a single-laser flow cytometer with two fluorescence channels of different receptor wavelength regions. F1 is excited from a labeled reference cell, using a laser beam able to excite both fluorescences and the photomultiplier tube (PMT) voltage of the channel for F2 is adjusted so that no signal is detected from F1. The test cell, labeled with both F1 and F2, is then excited and emissions from F1 and F2 detected in separate channels.
