High pressure liquid chromatography (HPLC) column for investigating the affinity of test substances (I) for melanin has a stationary phase (column filler material) consisting of silica gel to which eumelanin (EM) or phaeomelanin (PM) is covalently bonded. The EM is synthesised in vitro from L-dihydroxyphenylalanine (L-DOPA); and the PM is synthesised in vitro from L-DOPA and cysteine. The chemical bonding method also allows the use of melanins of biological origin, e.g. the melanin from the retina. For characterising the binding behaviour (corresponding to the affinity) of (I) with melanin, a defined amount of (I) is applied to the column and the corresponding retention is determined by isocratic elution. This retention depends on the amount applied and characterises the affinity of (I). The specific volume of the column is determined by applying sodium maleate having no affinity for melanin after each run.
