The following are claimed: (A) use of the method described in the specification (see below) for detection of, and determining the concentration of, pyrophosphates, polyphosphates and other inorganic polyanions which are from natural sources or are synthetically produced; (B) use of the method described in the specification for detection of, and determining the concentration of, compounds (which are from natural sources or are synthetically produced) which have multiple negatively charged groups and which are capable of selective and high affinity binding to certain polycationic compounds; (C) use of the method described in the specification for detection of, and determining the activity of, enzymes that catalyse reactions involving: (i) pyrophosphates, polyphosphates and other inorganic polyanions which are from natural sources or are synthetically produced or (ii) compounds as described in (B); (D) use of the method described in the specification for measurement of non-enzymatic reactions of compounds described in (i) and (ii) above, and (E) use of the method described in the specification for differentiation between linear and cyclic polyphosphates. N.B. No precise definition of the method of the invention is given. Broadly, it is based on the known fact that certain metal ions can quench or enhance the fluorescence intensity of cation-sensitive fluorescence dyes. Thus, the fluorescence intensities of fura-2, fura-Red, calcein and mag-fura-red are almost totally quenched by metal ions such as Mn2+, Co2+, Ni2+, Fe2+, Cu2+, Hg2+, Tb3+, Cu+, Zn2+ and Cd2+, and the fluorescence intensities of calcium-green-1, calcium crimson and fluo-3 are enhanced by metal ions such as Cd2+, Ca2+, Hg2+, Tb3+ and Zn2+. As polyphosphates, pyrophosphates and other similar ions show a high affinity to metal ions (especially di-cations), these compete with the above dyes for binding of the metal cations. The primary effect of these cations on the dyes is thus affected by the presence of increasing concentrations of the polyphosphates, pyrophosphates or other similar ions. This effect may be used for detection and determination of these ions.
