Thrombin-based assays are disclosed for determiningantithrombin III (ATIII) present in a plasma sample. Theassays involve a heparin derivative which, like heparin,effectively enhances the antithrombin activity of ATIII,but which, unlike heparin, does not substantially enhancethe antithrombin activity of Heparin cofactor II (HCII).In the present assay, HCII contributes about 15% or lessto the determined antithrombin activity. A lyophilizedreagent composition, useful in the ATIII assay andccmprising thrombin and a heparin derivative, oralternatively comprising thrombin and heparin, is alsodisclosed. A high-calibrator plasma reference useful inthe ATIII assays and in assays for other plasmaconstituents is also disclosed. Kits employing theseveral aspects of the invention are disclosed as well.
