SUBSTANCE: strain of bacterium Bacillus badius ВКМП B-6616 provides preparing restrictase Bba 179 I which recognizes and splits the nucleotide sequence 5′-(A/T)CCGG(A/T)-3′,. Strain does not show demanding to nutrient media, cultivation period is short. Strain B. badius ВКМП B-6616 is isolated from the soil as a result of the search of restrictase producers. The yield of enzyme is 1000 U/g crude biomass, specific activity of the end enzyme is 3000 U/ml. Restrictase Bba 179 I is an isoschizomer of restrictase Bet I. EFFECT: preparing restriction endonuclease indicated above, increased yield of enzyme. 1 dwg
