PURPOSE: To provide a method for simultaneously detecting the synthesized amount of repair DNA of a cell and the synthesized amount of replicative DNA, capable of being simply and rapidly carried out and measuring carcinogenicity of a chemical substance. ;CONSTITUTION: A first group in which a hepatocyte is floated in a culture solution and the number of cells is adjusted is mixed with a culture solution not containing hydroxyurea (HU) and radioisotope (RI) and cultured. A second group is mixed with a HU-containing culture solution and RI and cultured. A third group is mixed with the culture solution not containing HU, RI and a chemical substance specimen and cultured. A fourth group is the HU-containing culture solution, RI and the chemical substance specimen. After the culture, the culture solution is removed, RI not taken in a deoxyribonucleic acid (DNA) is removed. Then the DNA is made into a naked nucleus and purified. The total synthesized amount of each DNA-protein complex is measured for the first to fourth DNA-protein complexes derived from the first to fourth groups obtained by the DNA purification to calculate simultaneous equations formed between the DNA-protein complexes.;COPYRIGHT: (C)1995,JPO
