Highly-sensitive analysis of bile acids (BA) comprises reacting (BA)-contg sample with a reagent (R) consisting of (1) steroid dehydrogenase (SH) which catalyses reversible conversion of (BA) to oxo-bile acid (OBA), such conversions requiring as coenzymes (i) thio-NAD(P) or analogues and (ii) NAD(P) or analogues; (2) a cpd (A1) and (3) a cpd (B1). (A1) is a thio-NAD(P) or NAD(P) cpd in oxidised form; (A2) is its reduced form; B1 is reduced NAD(P) when (A1) is thio-(NAD(P), or reduced thio-NAD(P) when (A1) is NAD(P); (B2) is the oxidised form of (B1). (R) may include (a) a second dehydrogenase (H2) which does not react with (BA) but catalyses conversion of (B2) to (B1), plus a substrate for H2; in this case the amt of (A2) formed is measured; of (b) a third dehydrogenase (H3) which does not react with (BA) but catalyses conversion of (A2) to (A1), plus a substrate for H3; in this case the amt of (B1) consumed is measured. USE/ADVANTAGE - The method is esp used to assay (BA) in serum etc for clinical diagnosis of liver function, eg to establish the cause of jaundice, cholelithiasis, etc. The use of thio-NAD(P) as one coenzyme provides a more sensitive assay.
