Proteins isolated from crude extracts of the medicinal leech (Hirudo medicinalis) have been shown to strongly inhibit platelet aggregation and platelet aggregation and thrombogenous continuous activity of platelets in collagen. In addition, the protein prevents binding of von Willebrand factor to collagen. Methods for their use for blocking collagen-stimulated platelet aggregation as well as isolation and purification of such proteins are described. The novel protein (brandinin) has a molecular weight of approximately 15 kD and binds to collagen but has no activity to cleave collagen. The proteins are useful for preventing and treating thrombotic diseases and for coating blood-contacting materials that have a thromboresistant property.
