PURPOSE:To obtain the titled DNA useful for efficiently producing pyruvic oxidase capable of catalyazing reaction for producing acetyl phosphate, carbon dioxide and hydrogen peroxide from pyruvic acid, phosphoric acid and oxygen by applying a genetic engineering technique. CONSTITUTION:A microorganism, e.g. Lactobacillus delbrucki or Streptococcus sp., having the ability to produce pyruvic oxidase is cultivated in a liquid culture medium by a spinner culture method with aeration and the resultant culture is centrifuged to collect the microorganism. The resultant microorganism is bacteriolyzed to prepare a bacteriolytic material containing pyruvic oxidase. DNA is then separated, purified from the above-mentioned bacteriolytic material and cleaved with a restriction enzyme, e.g. EcoRI or HindIII, to provide a DNA fragment, which is linked to a vector fragment by a method for using a well-known DNA ligase to afford a recombinant DNA. The obtained recombinant DNA is then introduced into a host microorganism e.g. belonging to Escherichia coli, to provide a transformant, which is then cultivated to afford the aimed pyruvic oxidase.
