PURPOSE:To remove the cell wall of a lypase-producing microorganism effectively, and to form the protoplast of the above microorganism, easily and economically, by using one or more kinds of cell-wall lypases such as cellulase, pectinase, etc., in the preparation of the protoplast of the above microorganism. CONSTITUTION:A lipase-producing microorganism belonging to Candida genus, such as Candida paralypolytica, etc., is cultured in a potato sucrose liquid medium at about room temperature under shaking. In the above cultivation, the microbial cell is treated with the above cell-wall lypase in a weakly acidic or neutral buffer solution containing an osmotic pressure regulator by conventional method at 20-30 deg.C for 1-24hr to obtain protoplast and/or spheroplast. Since the cell wall of the lypase-producing microorganism is removed by the enzymatic treatment, the extraction and separation of the lypass in the cell can be carried out extremely easily. Namely, the cell can be disintegrated by simply dispersing the cells in a low-tonic liquid such as water.
