A method for performing at least two successive reactions, the product of the first reaction acting as a substrate for the subsequent reaction, applicable to DNA and RNA amplification techniques and avoiding contamination risks. In the method for performing at least two steps of a molecular biology experiment in a single Eppendorf tube, an amount of a first reaction medium (A) is placed in the tube, the medium (A) is covered with oil, a second reaction medium (B) is placed directly onto the oil, the media (A) and (B) are mixed after the reaction has been carried out in medium (A) and/or (B), and a reaction is incubated in the resulting mixture of A + B while using the products of said reaction(s) as a substrate. The various steps may be carried out without opening the tube.
