RECOMBINANT CHERRY SALMON GROWTH HORMONE, ITS STRUCTURAL GENE, RECOMBINANT PLASMID, BACTERIUM CONTAINING THE SAME PLASMID AND PRODUCTION OF GROWTH HORMONE OR PREGROWTH HORMONE OF THE SAME SALMON
PURPOSE: To readily mass produce the title substance useful for study of physiological action of growth hormone by culturing a microorganism containing a plasmid integrated with a gene of a recombinant growth hormone of cherry salmon derived from mRNA of pituitary gland of cherry salmon. ;CONSTITUTION: mRNA secreted from pituitary gland of cherry salmon is isolated by guanidium thiocyanate method, cDNA is synthesized by using the mRNA as a template, treated with a restriction enzyme and linked to a plasmid to prepare a plasmid library, which is transduced to Escherichia coli. The Escherichia coli is transformed and cultured and about 1,200 colonies are immobilized. A positive clone containing a gene encoding growth hormone of cherry salmon is selected by using a probe prepared by labeling a fragment of cDNA of pregrowth hormone with an enzyme, a microorganism containing a recombinant plasmid integrated with the gene is cultured in a culture medium and a product in the culture medium is separated and purified to give the objective recombinant cherry salmon growth hormone wholly or partially containing an amino acid sequence of the formula.;COPYRIGHT: (C)1993,JPO&Japio
