PURPOSE: To obtain a novel method useful for immune expression classification containing direct label absorbing staining and a reagent compsn. therefor. ;CONSTITUTION: Bovine intestine alkaline phosphatase is combined with a mouse monoclonal antibody (anti-CD2, CD3, CD4, CD8, CD19) for the purpose of direct immune enzymatic labelling. An erythrocyte is pre-prepared for hemolysis by ammonium sulface in a first culture period together with the alkaline phosphatase combined monoclonal antibody and, subsequently, a leucocyte is fixed early in the presence of dextrose. A combination of 5-bromo-4-chloro-3- indolylphosphate and nitroblue-tetrazolium salt in a diethanolamine buffering agent containing zinc glycine is used as a buffering agent/substrate under a pH condition of 9.5±0.1 in order to generate a stable and insoluble strong purple precipitate on the surface of a cell labelled with the alkaline phosphatase combined monoclonal antibody.
