This biosensor comprises a membrane 8 in which is immobilized an enzyme catalyzing the transformation of a substrate involved in the skin metabolism, means for detecting and measuring a phenomenon caused by this transformation, representative of the concentration of the substrate to be assayed brought into contact with the membrane 8 and a measurement cell 13, a wall area of which is formed by the membrane 8, and which is capable of being closed, during the measurement, by a skin covering area 15, the cell 13 being associated with means for circulating therein a liquid for dissolving the substrate to be dosed. For the measurement, the biosensor is applied to the skin, buffer is injected into the cell and once the latter is filled, detection and measurement are carried out, then buffer is again circulated to remove the solubilized substrate. This measurement cycle can be repeated without removing electrode 1. Application to the determination of cutaneous L-lactate by a Clark electrode associated with an L-lactate oxidase membrane.
