NEW MATERIAL:A type-specific DNA probe of herpes simplex virus containing a DNA of herpes simplex virus type 1 or 2 labeled by nick translation process. USE:Diagnostic for infection with herpes simplex type 1 or 2. PREPARATION:DNA extracted from a tissue-culture cell of a herpes simplex virus type 1 or 2 is used as a prototype DNA to obtain a clone of said DNA fragment. The obtained clone is screened by a colony hybridization using a type-specific DNA probe of a herpes simplex virus containing the DNA of herpes simplex virus type 1 or 2 labeled by nick translation process. A vector (plasmid) DNA is separated from the obtained clone and a DNA fragment is prepared therefrom. The fragment is purified by Southern hybridization technique to obtain a purified-type specific DNA probe for herpes simplex virus. The purified probe is screened by the hybridization with a wild strain DNA of herpes simplex virus.
