PURPOSE:To improve the elucidation of an expression mechanism in a facial cleft embryonic period and the research of the identification of an gene for the facial cleft disease by mating and raising rats so as to maintain the line of microphthalmia rats expressing the facial cleft. CONSTITUTION:Three lines of microphthalmia rats obtained in the first generation are independently subjected to brother-sister mating processes repeated =20 generation times and the expressing rate of the facial cleft is now kept constant. Any hybridization will be hereinafter not carried out between the three lines, and each line will be regarded as a closed colony and subjected to a closed circulation mating process. The mating is always performed between the male and female rats of the microphthalmia and rats expressing characters except the specific character are removed. The raising of the rats is performed in a breeding chamber employing a barrier system at a temperature of 23+ or -2 deg.C and at a humidity of 55+ or -10% under an artificial lighting for 13hr per day while feeding sterilized feeds and drinking water.
