The invention relates to a method of selectively hydrolysing organized phospholipids by means of lipolytic enzymes having regulated penetrating and hydrolising capacity due to site-specific modification with a hydrophobic group. More particularly, useful site-specific acylated pancreatic phospholipases A2 are capable of hydrolysing phospholipids of organized lipid-water interfaces, for example micelles, vesicles, liposomes and other bilayer structures, hexagonal HII-phases or biological membranes. The invention also relates to a method for the preparation of these modified enzymes and to the enzymes in substantially pure form.
