14A b s t r a c tLiability in human individuals to develop non-insulin-dependent diabetes mellitus (NIDDM) and/or athero-sclerosis is determined by restriction enzyme mappingof DNA from a human individual using a probe selectedfrom the group consisting of(i) cDNA complementary to the mRNA coding forthe human insulin,(ii) human genomic DNA containing the actual insulingene,(iii) DNA sequences of human genomic DNA locatedwithin 20 x 106 base pairs from the insulingene in either direction,and examining the distribution of DNA fragments forthe appearance of insertion sequences of approximately1600 to 2300 base pairs (U alleles) the occurence ofwhich indicates a liability for said individual todevelop elevated blood glucose concentrations and/oratherosclerosis and in homozygous form a liability todevelop NIDDM.
