Disclosed is a new enzyme-free glucose sensor applied to an in-situ test in a fermentation process. The sensor can test the concentration of glucose in a fermentation liquor. The present invention comprises the following content: preparing an Au@Ni/C nano-composite with a core-shell structure by using a high-temperature oleylamine reduction method, and according to this, preparing a working electrode; constructing a three-electrode test system; three electrodes capturing an electrical signal generated by the working electrode performing electrocatalytic oxidation on glucose; obtaining data corresponding to the concentration of the glucose; and carrying out calculation to obtain the concentration of the glucose. Compared with existing enzyme glucose biosensors, the sensor in the present invention does not need complex immobilization technology, is resistant to high temperatures, can be used for an in-situ test, is high in precision, has catalytic activity not influenced by fermentation environments such as temperature and humidity, has good stability, and can be applied to a long-term fermentation process on-line test environment. Compared with existing enzyme-free glucose biosensors, the sensor in the present invention works under a relatively low working voltage, is high in selectivity, is not influenced by adsorption of a large number of chloride ions present in a fermentation liquor, and is highly anti-toxic. Therefore, the sensor becomes an enzyme-free electrochemical sensor applied to an in-situ test of glucose in a fermentation process.
