Disclosed are a high-throughput mass spectrometry detection kit and a quality inspection method therefor. The kit comprises a pore plate, a biological sample pre-treatment material, a mass spectrometry test reagent and a quality control sample, wherein the biological sample pre-treatment material is 0.03-0.05 mg of porous silicon particles; and the mass spectrometry test reagent comprises an incubation solution, a cleaning solution, a matrix dissolving solution and a matrix. By calculating the average distance DL value between a detection sample and a quality inspection standard database and performing two-dimensional measurement on a mass spectrum by means of combining a cityblock distance and a Minkowski distance, the quality inspection method can improve the identification accuracy; the Youden's index of the identification of various variants is higher than 98%; and by judging the quality inspection qualification rate by means of evaluating the similarity of mass spectra, the quality inspection method can effectively control the whole production process of the kit, and the production process stability and the product stability can be evaluated. Therefore, the high repeatability of a mass spectrum reagent in a biological sample detection process is achieved, and a variable coefficient is controlled within 15%.
