The present invention relates to a method of manufacturing a microwell array having a hydrophobic-hydrophilic surface, and the present invention relates to a hydrophilic chamber for culturing microorganisms formed by combining a base and a partition wall, and a hydrophilic chamber formed in the partition wall and connected to the chamber and preparing a microwell array in which a plurality of microwells including an injection hole having a cross-sectional area smaller than the cross-sectional area of the chamber are formed; applying a hydrophobic material to the exposed surface of the partition wall and the inner peripheral surface of the injection hole so that the outside of the microwell is more hydrophobic than the inside; and curing the hydrophobic material so that the exposed surface of the partition wall and the inner circumferential surface of the injection hole are hydrophobic, whereby the injection hole of the microwell is formed with a hydrophobic surface and the chamber of the microwell is formed with a hydrophilic surface. It provides a method of manufacturing a microwell array having a hydrophobic-hydrophilic surface, comprising the step of fabricating the array. According to the present invention, first, microbial suspension injected into the microwell can be loaded more quickly due to the difference in affinity with water (hydrophobicity-hydrophilicity) between the inside and outside of the microwell, as well as the narrowed structure of the microwell and the inlet. The microbial suspension loaded into each microwell can be clearly partitioned from the microbial suspension loaded into the adjacent microwell by the surrounding hydrophobicity.
