摘要

以玉米胚芽为原料,用碱溶液浸提原料,将浸提后的溶液离心取上清液,调pH至蛋白等电点后继续离心,所得沉淀即为玉米胚芽蛋白.以碱浓度、酸沉pH、料液比为考察条件,以玉米胚芽蛋白提取率为指标进行研究,经单因素初步摸索后,建立三因素三水平编码,碱浓度水平条件分别为0.2%、0.3%、0.4%,酸沉pH水平条件分别为4、4.5、5,料液比水平条件分别1∶5、1∶10、1∶15,进行响应面实验.将正交实验结果进行方差分析并建立描述蛋白提取率的回归模型,分析结果表明,F差异不显著,可认为所选的二次回归正交模型是合适的.对玉米胚芽蛋白提取率影响大小的顺序为碱浓度>料液比>酸沉pH.两因素的交互作用中,碱浓度与料液比对蛋白提取率影响显著.由正交试验得到的结果,玉米胚芽蛋白提取的最优条件为NaOH浓度0.3%,蛋白等电点4.5,料液比为1∶10.%Corn Germ was used as raw material,liquid was acquired after alkaline solution soaked the material by centrifugation.Centrifugate again when the liquid was adjusted to the protein isoelectric point,the obtained precipitate was corn germ alkali-soluble protein.Alkali concentration,pH and solidliquid ratio were considered as factors,protein extraction yield was employed as evaluation index.Response surface test was carried out with alkali concentration was 0.2%,0.3%,0.4%,pH was 4,4.5,5 and solid-liquid ratio was 1 ∶ 5,1 ∶ 10,1 ∶ 15 respectively.The results of orthogonal experiment was of variance analysis and a regression model was established to describe the protein extraction rate.
