The studies on decolourisation of distillery effluent can not be carried out efficiently without the knowledge of the nature of colourant(s) and their properties. The presence of brown colour in distillery effluent is mainly due to the coloured compounds, which are formed during manufacturing of sugar from sugarcane juice. Caramels and melanoidins constitute the main colouring ingredients, which are formed by non-enzymatic reactions. In this study, one of the colouring compounds from the distillery effluent was purified by thin layer chromatography and adsorption chromatography. The structure of the purified coloured compound was elucidated by the application of the spectroscopic techniques, viz., UV, NMR, IR and Mass Spectoscopic analysis. The compound was characterized as 3, 4 dimethoxy 2 (1 hydroxy ethyl) α-naphthol. Biotransformation of the brown colouring compound was carried out by the treatment of predigested distillery effluent with indigenous bacterial consortia of activated sludge. The two colourless products formed after biotransformation were purified and characterized by UV, NMR, IR and MS analysis. The interpretation of the spectroscopic analysis led to the conclusion that the two products formed after biotransformation were 3, 4 dihydroxy α-tetralone (1) and 4 - hydroxy α-tetralone
