Antigenic and genetic diversity of Orientia tsutsugamushi among strains is well demonstrated by serological tests with polyclonal or monoclonal anti-bodies and by PCR-RFLP analyses of type-specific 56-kDa antigen gene. Most recent isolates from patients in Japan were classified into 4 types, JG, JP-2, Kawasaki and Kuroki, but infections of Kato and Shimokoshi types were found sometimes. Such antigenic variation depends on the diversities of an immunodominant 56-kDa antigen located on surface of the microorgansim. To further clarify the relationship between individual strains, the 56-kDa gene of each srain was amplified by PCR and sequenced in an automated nucleotide sequencer (Pharmacia), and the relationship among strains was determined by phylogenetic analysis based on the sequence homologies in the present study. Strains used in this analysis were 6 prototype strains of Gilliam, Karp, Kato, Kawasaki, Kuroki and Shimokoshi, and 50 isolates from patients, natural rodents and mites in Japan, South Korea, China, and Thailand.
