Photoacoustic tomography has attracted high attention in recent years, because this imaging technique can combine the advantages of ultrasound and light to improve the image resolution and diagnostic depth of tissues. In this paper, spectrospopic feature of photo-induced acoustic signal generated in biological tissues are investigated, and its characteristics are analyzed in time domain and in frequency domain. The results show that spectrum of photoacoustic signal changes with the property of tissue sensitively. A spectrum-selective photoacoustic imaging might be a potential approach to diagnose special disease at the early stage. According to the spectral characteristics of photoacoustic signal, here, we present a photoacoustic tomography of real tissue to depth of ~12mm by selecting a wide-band high-sensitive detector. The time-resolved technique and a phase-controlled focus algorithm was used for image reconstruction, and a depth resolution up to ~10 μm and a lateral resolution about 300 μm were obtained.
