摘要

This paper describes a verstile sensing membrane for immobilisation of proteins (antibodies, antigens, enzymes and so on) and a new immobolization method. The membrane was developed with poiyacrylaminde gel and biotin-streptavidin system. Streptavidin was bend to the polyacryamide gel with i-ethyl-3- (3-dimethylaminopropyl carbodimide (EDAC), which reaofts in a streptavidin membrane, and a biotinalyted ligand was then easily linked to the gel via the streptavidin molecules. The streptavidin membrane ( the thickness of 600nm) was used to immobilise anti-human chorionic gonadotrophin antibody (α hCG). The number of αhCG in the membrane was about 10~(11) per mm~2 of the membrane surface. The membrane was used as a sensing membrane of optical imsunosensor ( Mach-Zehnder interferomter). It had a good response at the concentration of 10~(-11)mol/L hCG.
