The PHFT machine is capable of performing polymer chain reaction, genechip hybridization and fluorescence detection automatically. There are fourteen tubes in the new biochip and the temperature in each tube can be changed quickly. Every tube contains a glass tube, a cell and a piece of glass. A aminosilane was used to modify glass surface and it was successfully used for attaching prefabricated DNA of oligonucleotides. The experiment showed that the PCR hybridization chip could distinguish an internal single base mismatch in 20mer oligonucleotides. The probes designed to detect the hepatitis B virus and some of its mutations were immobilized on a glass. The genome of hepatitis B virus in blood is separated and amplified and fluorescence labeled. After labeling, PCR products were hybridized to those probes; the PHFT instrument could detect the fluorescence image and calculate the fluorescence intensity. The results indicate the new biochip will find application in clinical HBV and its mutation detection.
