Stir bar sorptive extraction (SBSE) coupled with thermal desorption unit (TDU)-gas chromatography–mass spectrometry (GC–MS) (SBSE-TDU-GC-MS) was applied to detect poly aromatic hydrocarbon (PAH) especially 1-Naphthylamine (1-NA) and 2-naphthylamine (2-NA) in 138 human urine samples. A stir bar coated with polydimethylsiloxane (PDMS) is added to a 1ml urine sample and stirring is carried out for 60 min at room temperature (25 ℃) in a vial. Then, the PDMS stir bar is subjected to TDU–GC–MS. Urine samples without and with enzymaticde-conjugation were analyzed. Results show that, in the urine samples without enzymaticde-conjugation, 1-NA was detected in 58 samples. In contrast, in the samples treated with (β)-glucuronidase, it was detected in 90 samples. These results indicate that most of 1-NA and 2-NA presented as conjugate in the urine samples. In addition, it is found that 1-NA and 2-NA presence is closely related to the lifestyle of the people and it is independent on gender, age and body-mass index (BMI).
