Isolation, partial identification and copper tolerance of some native species of the three major groups of iron and/or sulfur oxidizing prokaryotes (mesophiles, moderate and extreme thermophiles) from the Sarcheshmeh Copper Mine are described. There are vast reserves of low grade copper ores (about 280,000,000 tones) in this mine which the NICICo (National Iranian Copper Industries Company) plans to bioleach it, therefore it was considered to elucidate the Mine's native microflora, and then to determine their tolerance of leaching conditions, such as low pH values, high metal concentrations and pulp densities. Samples were taken from certain parts of the mine and cultured in defined liquid media. Representative strains of all three temperature-related groups used in bioleaching were isolated from which some purified cultures were obtained. These were a strain of Thiobacillus ferrooxidans (a mesophile), an endospore forming moderate thermophile and an extremely thermophilic isolate. Following this, attempts were made to enhance copper tolerance of some isolated strains. A Thiobacillus sp. increased its resistance from 5 to 20 g/1 copper in the culture media. The results showed that all the important bacteria used for bioleaching, are present in Sarcheshmeh Copper Mine.
