Continuous monitoring of in vivo human brown adipose tissue in response to cold stimulation by photoacoustic tomography

The metabolic activities of brown adipose tissue (BAT) between healthy and diabetic or obese population are different when they are exposed to cold stimulation. In this study an experimental platform based on label-free photoacoustic tomography (PAT) was designed to in vivo monitor the development of BAT over time in human supraclavicular area in response to cold exposure in near real time. Specifically, 7 healthy volunteers were recruited and monitored in vivo by the PAT system at the wavelength of 800nm continuously for 420 s, where haemoglobin acted as the endogenous contrast agent. The PAT images obtained showed that the mean photoacoustic signal intensity of the BAT increased rapidly after cold stimulus, while the photoacoustic signal of the blood vessels in the supraclavicular region decreased upon the cold stimulation. To the best of our knowledge, this is the first-in-human study to apply PAT for measuring the metabolic activity of BAT continuously.
