Controlled release paediatric paracetamol suppositories using non-melting, non-disintegrating hydrophilic matrices have been developed. The formulations showed matrix diffusion controlled drug release with initial fast release providing for the loading dose followed by slow release of maintenance dose. In vivo evaluation in rabbits showed therapeutic levels maintained till 8 h as compared to 4 h with marketed product.
