A series of in situ forming injectable hydrogels based on poly(L-glutamic acid) and alginate were designed without incorporating any extraneous cross-linking agents. The gelation time, equilibrium swelling, storage modulus, microscopic morphology could be adjusted by varying the solid content of the hydrogels and oxidation degree of alginate. ASCs incorporated in the hydrogels remain viable. A preliminary in vivo study exhibits injectability and rapid in vivo gel formation, as well as mechanical stability, cell ingrowth and adipogenic differentiation. These studies indicate that the PLGA/ALG injectable hydrogels may have potential uses in adipose regeneration and other soft tissue engineering applications.
