Conclusions: This in vivo study suggests that NF-κB decoy ODN treatment mitigates wear particle-associated osteolysis using the murine femoral particle infusion model. Decoy ODN treatment increased BMD in femurs exposed to the particles, but had no significant effects on systemic macrophage recruitment. This suggests that the therapeutic effects of decoy ODN are mediated by a reduction of macrophage-associated inflammation and osteoclast activation in the region of particle infusion and drug delivery, rather than systemic macrophage chemotaxis.
