THE ANTINOCICEPTIVE EFFICACY OF CHRONIC FLUOXETINE TREATMENT

Fluoxetine, a selective serotonin reuptake inhibitor, is one of the oldest second-generation antidepressants on the market. Recentstudies have shown that fluoxetine has antinociceptive effects, but the results were based on acute doses of the drug. Theobjective of our study was to compare the antinociceptive efficacy of two fluoxetine doses delivered acutely and chronicallyto a naïve control using adult male Sprague-Dawley rats and a model of acute thermal pain involving a hot plate analgesiometer(Omnitech Electronics, Inc.; Columbus, OH) set at 52.5°C. The treatment groups received 10 mg/kg or 20mg/kg of fluoxetinethat was delivered daily for two weeks or four months via Pillsbury sugar cookie dough. The naïve control group receivednothing. Each animal was habituated to the hot plate enclosure for one minute on the day prior to testing. For the study, eachrat was placed on the hot plate apparatus, and a built-in timer was started as soon as all four of the animal’s paws touched thesurface. The timer was stopped as soon as the animal displayed one of the following responses: paw lift, paw lick, or jump.Our results show that both chronic fluoxetine treatment groups had significantly longer latencies compared to the naïve controlgroup, and that the latencies between the two doses were not significantly different. Overall, our results indicate that long-termadministration of fluoxetine via cookie dough is capable of reducing the response to thermal stimuli.
