We have rationally designed a hydrogel to mimic the invasive GBM tumour microenvironment. Using a screen of different ECM-mimetic peptides, we found that a fibronectin-mimetic peptide sequence promotes GBM and control human fetal cell adhesion on 2D TCP as well as in 3D hydrogels. We have demonstrated invasion of two GBM lines benchmarked to non-invasive human fetal neural stem cell lines. This work demonstrates a potential platform for probing mechanisms involved in cellular invasion in vitro as well as a potential platform for high-content drug screening against targets that may inhibit cellular invasion.
