The micropropagation system has been successfully developed for gerbera. Picking out the appropriate medium of the rooting culture is the key. In the test, micropropagated shoots of gerbera were cultured from capitula. Plantlets 4cm long with 3-4 leaves were rooted on half-strength Murashige and Skoog medium, supplemented with different types and concentrations of auxins, so as to determine the effects of composition and concentration of auxins on in vitro rooting and in vivo establishment of the gerbera plantlets. Among the various auxins tested, the best rooting response was obtained with 0.2-0.5 mg-L-1 indoleacetic acid (IAA), or 0.01-0.02 mg-L-1 a-naphthaleneacetic acid (NAA), or 0.05-0.5 mg-L-1 indolebutyric acid (IBA), both in terms of rooting frequency and survival rate after being transplanted. An increase in the concentration of auxins on in vitro rooting has detrimental influence upon survival rate of plantlets during acclimatization.
