This study concerns the development and evaluation of a new and more sensitive methodology for the estimation of milk (from goat, sheep and cow) content in Halloumi cheese, based on the detection of species specific mitochondrial cytochrome b DNA by means of Real Time Polymerase Chain Reaction (RT-PCR) and of fluorescence quantitative RT-PCR in order to increase the sensitivity of the quantitative analysis of the composition of milk in Halloumi cheese. The methodology is based on using three species specific and one common primer (control) that enable the amplification of different size DNA fragments for each species at the same time. The study began with the extraction of DNA from blood cells of goats, sheep and cows, which are bread in Cyprus for the production of dairy products, in order to confirm that the mitochondrial cytochrome b DNA fragments amplified by PCR can be used for the qualitative and quantitative analysis of species specific origin in milk solutions. The methodology under development with regard to the elution of total DNA from samples of raw and pasteurized milk of different animal origin and from samples of milk mixtures and milk products with various composition, will also be discussed with regard to correlations between species specific DNA recovered and the contents of milk used in the original product.
