High-throughput sequencing (HTS) techniques are revolutionizing biomedical sciences and have the same potential for environmental sciences. A proof-of-concept pilot project was undertaken to determine the efficacy of HTS to monitor the evolution of a microbial community in a tetracholorethene-contaminated aquifer undergoing remediation by enhanced reductive dechlorination (ERD). The selected groundwater remedy underway at the North Railroad Avenue Plume (NRAP) Superfund Site in Espa?ola New, Mexico (NPL #NMD986670156) is ERD using carbon source and nutrient addition. This aquifer is the sole source of potable water for Espa?ola and the adjacent Santa Clara Pueblo, but contamination from the NRAP source, a commercial drycleaner, forced the closure of two water supply wells. Prior to remedialaction, the NRAP groundwater plume comprised an approximate 58-acre, 260-foot-deep plume of tetrachloroethene, which is classified as a possible carcinogen.
