PACLIMER~TM microspheres is an injectable biodegradable polymer formulation of paclitaxel developed by Guilford Pharmaceuticals, Inc. Unlike traditional systemic chemotherapeutic regimes, PACLIMER~TM microspheres is designed to provide site-specific and controlled delivery of paclitaxel, and has been effective against various types of cancer in preclinical animal models~(1-2). In this paper, we report the preparation and in vitro characterization of paclitaxel microsphere formulation (PACLIMER~TM microspheres) using a robust reversed phase HPLC method for quantitation and further structural elucidation using LC/MS techniques. The polymer used in the microsphere formulation is a biodegradable polyphosphoester (PPE) polilactofate polymer, which has shown great promise in controlled delivery of a variety of therapeutic agents, ranging from small molecules to proteins and nucleic acids~(3-5).
