Control of Haemophilus parasuis infections depends on the accurate diagnosis of the agent, identification of the prevalent strains causing disease in the herd, management of the introduction of new strains into the herd and use of adequate vaccine programs. Diagnosis and identification of H. parasuis is usually based on the isolation of the organism from lesions and further characterization of isolates by serotyping and or genotyping. Isolation of H. parasuis from clinical samples has been reported as difficult and frequently unsuccessful. Unfortunately, all currently available typing methods for H. parasuis require the isolation and biochemical identification of the isolates. Recently a 16S rRNA gene-based PCR was developed to improve the diagnosisof H. parasuis infections. The PCR was able to detect the presence of H, parasuis in clinical samples even when isolation was not accomplished. Study of the restriction sites present within the PCR-amplified sequences from 12 H. parasuis reference strains demonstrated that these strains could be grouped into four PCR-RFLP types. In this study, we evaluated the use of PCR-RFLP as an alternative method for differentiation of H. parasuis isolates recovered from a field outbreak.
