摘要

A high-throughput, fully automated, multi-color fluorescent fingerprinting technique for large-insert genomic DNA clones was developed and used to fingerprint 215,645 genomic fragments of an Aegihps tauschii line that is genetically very closely related to the D genome of Chinese Spring wheat (Triticum aestivum). The fragments were cloned in BAC and BiBAC vectors. The fingerprints were edited with a computer program (GenoPro filer) we developed and assembled into contigs with the FPC program. A totalof 10,035 contigs were obtained at a Sulston score of 1><10"30 and a tolerance of 0.4 bp, corresponding to ca. ;200 Mb. Some of the contigs have been anchored to wheat maps by hybridization of RFLP markers and mapped ESTs with BAC and BiBAC clones. A database including fingerprints, BAC and BiBAC clones, contigs, and integrated markers has been developed and is publicly accessible at http: //wheatdb. ucdavis.edu: 8080/wheatdb/.
