Mutator, a wide-spread DNA-type transposable element, was first identified in maize and has been extensively characterized. To study Mutator-like transposable elements (MULEs) in wheat, homologous clones were isolated from Triticum urartu using degenerate primers constmcted according to the conserved transposase sequences of maize MuDR and its rice orthologue OsMu. DNA fragments encoding a part of the transposase were obtained by PCR. Deduced amino acid sequences of all 29 amplified DNA fragments with the expected length showed significant homology with the corresponding maize and rice transposase sequences. Nucleotide sequences were divided into two clusters using the UPGMA method. Some members in both clusters possessed intact transposase sequences without interrupting stop codons. DNA gel blot analysis was conducted in the two wild einkom wheat and several wild and cultivated polyploid wheat using representative sequences of the two clusters as probes. Hybridization pattems showed that one cluster consisted of low-copy elements, while the other consisted of remarkably high-copy elements. The results show that A genome of the wild einkom wheat possesses at least two classes of MULEs that had greatly different transposition activities.
