The human T cell leukemia virus-1 (HTLV-1) is a retrovirus which causes adult T cell leukemia (ATL) and neurological disorder, the tropical spastic paraparesis (HAM/TSP). The pathogenic factor is highly likely to be the viral protein Tax, which is a key regulator of viral replication. In addition to viral replication, Tax exerts 1) trans-activation and -repression of transcription through interaction with various transcription factors and co-activators, 2) dysregulation of cell cycle through inhibition of some tumor suppressor proteins, and 3) accumulation of mutation in cellular genes through suppressing DNA repair enzymes and checkpoint functions. These pleiotro-pic effects on various cellular targets seem to cooperate in promoting cell proliferation and fixation of genetic abnormalities, ultimately resulting in cell transformation and leukemogenesis.
